Anti-Obesity Activity of Ethanol Extract of Veronica peregrina L.

Abstract
Veronica peregrina L. is a plant that grows mostly in Asia and Europe and is commonly seen near paddy fields and streamsides. The present study aimed to evaluate the inhibitory effects of ethanol extracts of V. peregrina L. (EEVP) on adipocyte differentiation and adipogenesis using 3T3-L1 preadipocyte cell line. The results showed that treatment with EEVP suppressed the terminal differentiation of 3T3-L1 preadipocytes in a dose-dependent manner. The inhibitory effects of EEVP were confirmed by a decrease in the lipid droplet content through Oil Red O staining in the mature adipocytes. The concentration of cellular triglycerides were also reduced in 3T3-L1 cells with EEVP treatment when compared to the controls. EEVP inhibited adipogenesis and downregulated the expression of adipogenic transcription factors, such as peroxisome proliferator-activated receptor-¥ã sterol regulatory element-binding protein 1c, CCAAT/enhancer-binding protein ¥á and ¥â, and adipocyte expressed genes, such as adipocyte fatty acid binding protein and leptin. In addition, EEVP treatment effectively increased the phosphorylation of the AMP-activated protein kinase and acetyl CoA carboxylase. This study demonstrated that EEVP has a preeminent effects on the inhibition of adipogenesis, indicating its potential as an anti-obesity agents.
